It should. There's just two words of advice:

1) Make sure you're not using a venting regulator. These essentially include an overpressure valve in them and, placed in the pilot area like that is, could potentially trigger the valve. Needless to say, that's not safe.
2) You need to work out how you're loading the thing to make decent use of an on-board air supply.
Does that thing kinda look like a big cat to you?

Seriously though, you should use a three way valve instead of a blowgun.

one thing you could have a paintball tank attached to it and still have it regulated down lower to 30 or 40 psi, and i think some pvc will hold 200 psi.
when life gives you lemons... make root beer and leave the world wondering how you did that :D

About 15 shots at 20 psi output from a 100 psi main chamberererer.

37 cubic inches sounds far too big for shooting Nerf darts though, have you tried modelling your idea in GGDT?

I have not yet modelled my design yet, so I don't really know what the dimensions should be. i want to be able to move the gun around easy.

I don't know if there is other type of slide valve but with the common available one with that design you will be pushing the trigger than pull it. You should move the valve near the qev to have forward direction, except you want to add more mechanical to the trigger.
